Many rare wood Chiropterans live in the Park (Bechstein's bat, Greater Noctule bat, Noctule bat, Barbastelle), as well as some species of great preservation value, such as the Porcupine, the Polecat and the Red deer. The Squirrel has recently reappeared in the pinewoods near Ravenna.
Unfortunately some allochthonous species are present too, some of which endanger the wetlands original plants and animals (Coypu).
The Otter became extinct towards the middle of the '80s.

The Delta is the most important zone for birds in Italy and one of the most important in Europe.
For several species, the Delta represents a real stronghold at a national and European level, e.g. for Squacco heron, Purple heron, Ferruginous duck, Avocet, Black-winged stilt, Mediterranean gull, Gull-billed tern, Little tern.
Some species are internationally very rare, such as the Pygmy Cormorant - with the only colony in Western Europe - the Rüppells Warbler - with the only nesting couples of the whole continent - and the Flamingo, with one of the few European colonies. Also many other important species are worth mentioning: Bittern, Spoonbill, Glossy ibis, Shelduck, Red-crested pochard, Oystercatcher, Collared pratincole, Sandwich Tern and Whiskered Tern.
Among the nesting species, which have been recently reported, there are: Lesser kestrel, Red-footed falcon, Roller and Spanish sparrow.

Among the most interesting species there are the European pond turtle and Hermanns tortoise. The first is widespread in freshwater wetlands, while an autochthonous relict core of the second can be found in the wood Bosco della Mesola.

The Park houses three endemic species, one from Italy (Crested newt) and two from the Po valley (Italian Agile Frog, Italian Tree-Frog). The endemic Rana latastei (Italian Agile Frog) is very important, since it is very rare and lives exclusively in wet and flooded forests. In the Park it is localized only in Punte Alberete, the southern most distribution site.
The presence of the Pelobates Fuscus has been recently attested to.

In the Park there are ten endemic species: one Adriatic (Adriatic Sturgeon), two North Adriatic (Knipowitschia panizzae, Potamoschistus canestrinii), three from Italy (Laska Nase, Rutilus rubilio, Italian Barbel) and four from the Po valley (Rutilus erythrophthalmus, Chondrostoma soetta Sabanejewia larvata, Padogobius martensii).
The Adriatic Sturgeon and Sabanejewia larvata are the most important endemic species, since the first is extremely rare and the second, within the Park, is localized only in Punte Alberete, the southern most distribution site.
Among the other species there are: Aphanius fasciatus, Three-spined Stickleback, Lipophrys fluviatilis and the Common Eel, which is the symbol of the Park.

Only one species is present in the Park, it migrates from the sea to the streams where it lays eggs: the Sea Lamprey.
